Nagisa Misumi, son derece sportif ama çalışmaktan da o derece nefret eden biraz erkeksi bir kızdır. Honoka Yukishiro ise Nagisa'nın tam tersine zeki ve tam bir kitap kurdudur. Aynı okulda okuyan ancak birbirlerini yolda geçenken gören onun dışında hiçbir yakınlıkları olmayan bu iki kız, tüm dünyanın kaderini etkileyecek bir olay ile bir araya gelmek zorunda kalırlar.

Nagisa, eve döndüğünde cep telefonuna benzeyen bir alet ve kartlar bulur. İçinden çıkan Mippuru, Nagisa'nın kendisine Meppuru'yu bulması için yardımını ister. Ancak Meppuru çok yakınlarında, Honoka'nın yayındadır.

Mippuru ve Meppuru, "Dusk bölgesinin" hakimi; "Evil King", "Hikari no Sono"yu ele geçirdiğinde oradan kaçarak dünyaya gelmişlerdir. Mippuru ve Meppuru'nun kendilerine verdiği güç ile Nagisa; "Cure Black"a, Honoka ise "Cure White"a dönüşmüştür. Birlikte "Evil King" ve uşaklarının kötü planlarını durdurmak için savaşmaya başlarlar.
